Executive Summary

1. Established drug, transformed delivery: ONX-1006 builds on paclitaxel, whose anticancer activity is already proven in humans, and is designed to substantially improve its tumor delivery, tolerated exposure, and treatment duration.
2. Compelling preclinical evidence: 30+ studies established a NOAEL equivalent to 750+ mg PTX for a 60 kg adult, about 3.8 times a typical Abraxane dose.
3. Efficacy: strong antitumor activity in pancreatic cancer models, supporting development goals of 90–100% tumor elimination and 70% one-year survival.
4. Manufacturing: standardized process established with 15 kg successfully produced.
